Host Depletion

The presence of larger eukaryotic genomes can obfuscate sequencing signals from microbial genomes. The Devin Host Depletion filter is a proprietary syringe filter that effectively removes host background signals from all liquid biopsies. The patented technology, called Zwitterionic Interface Ultra Self-assemble Coating (ZISC), depletes host nucleated cells while allowing bacterial, viral, fungi, and parasite communities to pass through unaltered.

Enrichment and Purification

The Devin Microbial DNA Enrichment Kit is a fast and simple genomic nucleic acid purification protocol specifically designed using mNGS-grade reagents to minimize contamination. The kit begins with the Devin filter to deplete host nucleated cells from liquid biopsies. Extractions with the kit, after host depletion, are also compatible with automated systems, facilitating high-throughput NGS assays.

Library Preparation

The Unison Ultralow DNA NGS Library Preparation Kit optimizes the preparation of NGS libraries for low-input DNA (as little as 10 pg) extracts. The kit also accelerates DNA library preparations by utilizing a proprietary tagmentation-based method which condenses fragmentation, adapter ligation, and normalization steps into a single step. Specifically optimized for high molecular weight DNA the kit generates library fragments whose sizes are optimal for Illumina platforms at normalized concentrations.

Unison provides you with pure, high-quality DNA extracts that enable multiplexing each sequencing run. By effectively reducing host contamination right from the start, the sequencing depth required to achieve a representative microbiome sample can be reduced by several fold. This reduces sequencing costs and shortens overall sequencing time, depending on the downstream sequencing platforms.

To streamline the entire mNGS workflow, our PaRTI-Seq Analysis ensures fast reporting of results. The platform is curated with up-to-date genome information for thousands of microorganisms. The PaRTI-Seq Analysis pipeline has proper database benchmarking and robust analysis of microbial pathogens with unusual sequence homologies and/or close taxonomic relationships, reducing false-positive or false-negative results. Our PaRTI-Seq Analysis is available by the reaction or as an annual subscription.

PaRTI-Cular’s state-of-the-art computational algorithms provide a complete report within 30 minutes per sample. The cloud-based software also automatically conducts data quality checks, removal of host genome reads, pathogen identification, and statistical analyses.
