    Our research showcases a novel method for transforming low-yield, host-depleted DNA into high-quality, sequencible libraries using PCR amplification during library preparation on Oxford Nanopore Technologies (ONT) platforms. 

    • Host depletion resulted in a dramatic shift, reducing human reads from 97% to 14% and enriching microbial content from 0.34% to 85.96%.
    • PCR amplification successfully yielded 63ng/μl library DNA from host-depleted samples, a significant improvement over non-PCR-based methods, enabling efficient sequencing on ONT’s Flongle flow cells.
